Background

MUC1 is a large cell surface mucin glycoprotein expressed by most glandular and ductal epithelial cells and some hematopoietic cell lineages. It is expressed on most secretory epithelium, including mammary gland and some hematopoietic cells. It is expressed abundantly in lactating mammary glands and overexpressed abundantly in >90% breast carcinomas and metastases. Transgenic MUC1 has been shown to associate with all four cebB receptors and localize with erbB1 (EGFR) in lactating glands. The MUC1 gene contains seven exons and produces several different alternatively spliced variants. The major expressed form of MUC1 uses all seven exons and is a type 1 transmembrane protein with a large extracellular tandem repeat domain. The tandem repeat domain is highly O glycosylated and alterations in glycosylation have been shown in epithelial cancer cells.

  1. I would like to perform a sandwich assay for the determination of MUC1 protein. May you suggest me some antibodies that can be used together? In particular, if possible, I should need antibodies product in mouse or rabbit.
    • Unfortunately we are not aware of a pair of antibodies that have been used in sandwich ELISA in particular. If you are interested in trying two you may find our Innovators Reward Program to be helpful.
  2. We would like to perfrom ELISA to detect human MUC1. May we ask: 1. if NB120-10120 is suitable for coating and NB120-10119 for detection? 2. if #AR00024PU-N from Acris is suitable to work with NB120-10120 and NB120-10119, and be used as the standard? 3. your suggestion on other antibody combinations and standards?
    • It doesn't look like the lab has tested either of these together as a pair. Also they are both mouse monoclonal's so they would not be a suitable match as it would be best to have two antibodies of different host unless the customer plans on doing a direct conjugation. We recommend monoclonal's for capture and polyclonal's for detection. Most often unless specifically listed these our antibodies are not tested in sandwich elisa, but direct elisa. Most likely the Acris protein could be used as a standard, but we cannot say with 100% certainty as this was not tested with either of these two antibodies. It doesn't look like we have any recombinant protein standard that we offer that is specifically used with either of these two.
