Background

MSH6 is a mismatch-repair protein. It plays a role in tumor suppression. Genomic mutations in MSH6 predispose to the hereditary non-polyposis colorectal cancer (HNPCC) syndrome.

Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Western blot shows lysates of mouse liver tissue. Gels were loaded with 12 µg, 6.5 µg, and 3 µg of tissue lysate. PVDF membrane was probed with 1 µg/mL of Goat Anti-Mouse MMR/CD206 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2535) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for MMR/CD206 at approximately 180 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
MMR/CD206 was detected in perfusion fixed frozen sections of mouse lung using Goat Anti-Mouse MMR/CD206 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2535) at 25 µg/mL overnight at 4 °C.   1. I am testing a mouse sample with human xenograft for GBM and I am looking for an MSH6 Antibody that will only detect the human cells.
    • Currently, we have no validation that any of our MSH6 antibodies will not cross react with mouse. All of our MSH6 Antibodies are raised against Human MSH6 so we have checked the % homology with mouse MSH6 which has come to be 85%. As 85% is very high, the chances of the antibody detecting the mouse tissue as well as the human tissue are high.
