Background

IL-4 is a pleiotropic cytokine produced by activated T cells, mast cells, and basophils. IL-4 is a potent lymphoid cell growth factor which stimulates the growth and activation of certain B cells and T cells. IL-4 is important for regulation of T helper subset development. The 11B11 antibody reacts with mouse interleukin-4 (IL-4). The 11B11 antibody can neutralize the bioactivity of natural or recombinant IL-4.

1. One customer wanted to know the dilution of NB100-78173 for IHC-P,IHC-Fr, IF and WB. The datasheet haven't showed these information.
   • Clone 11B11 of this antibody is highly documented in research papers and the dilutions are little tricky as it depends mostly on the expression of IL4 in samples under testing. I have listed the suggested concentration range of this antibody below, however, your customer may go for lower or higher dilutions depending upon how much expression of IL4 is there in customer's particular samples. IHC-P,IHC-Fr: 5ug/ml-25ug/ml range. ICC/IF: 1ug/ml-5ug/ml WB: 1-5 ug/ml
2. Sorry to disturb you again for NB100-78173. The customer detected the protein expression of mouse's heart, liver, spleen, and kidney with IHC by your suggested dilution range, Unfortunately, the expression of IL4 was very low, so the customer wanted to know whether there are some tissue sample as positive control?
   • Because it is a secreted protein, I am not surprised that your customer is observing a low expression in tissues. Is your customer performing an induction experiment or looking at normal tissues? As far as tissues are concerned, spleen should be good for that. I can see some of our testing has been done on MCF-7 and HeLa cells as well. However, I would highly suggest your customer to use our IL4 Overexpression Lysate (H00003565-T01) as a positive control.
