Application Notes
Western blot analysis (1-2ug/ml), immunoprecipitation (4-8 ug/ml) and Immunohistochemistry (10-20 ug/ml). However, the optimal conditions should be determined individually. The antibody recognizes a 90 kDa protein, corresponding to the apparent molecular weight of Hsp90 on SDS-PAGE immunoblots. This antibody detects both Hsp90 alpha and beta.

Background

In mammalian cells there are at least two Hsp90 isoforms, Hsp90a and Hsp90b which are encoded by separate genes. These ubiquitous and highly conserved proteins account for 1-2% of all cellular proteins in most cells. Hsp90 is part of the cells powerful network of chaperones to fight the deleterious consequences of protein unfolding caused by nonphysiological conditions. However, in the absence of stress, Hsp90 is a necessary component of fundamental cellular processes such as hormone signaling and cell cycle control. In this context several key regulatory proteins such as steriod receptors, cell cycle kinases involved in signal transduction and p53 have been identified as substrates of Hsp90. It has been suggested that Hsp90 acts as a capacitor for morphological evolution by buffering widespread variation, which may affect morphogenic pathways.

  1. We were wondering how many western blots could be performed using your recommended dilutions and 0.05 mg of this antibody.
    • The number of Western Blots you will be able to perform with 0.05mg of antibody can vary greatly from product to product. We have a number of HSP90 antibodies to choose from, but one I see that is supplied as 0.05mg is NB120-1429. We recommend using 1-2ug per milliliter of diluent for this antibody. Unfortunately, I cannot give you an exact answer because it depends on the amount of diluent you will be using. Typically, our customer can get a good number of blots using our products.
