Immunohistochemistry-Paraffin: Notch-1 Antibody (mN1A) [NB100-78486] - Analysis of FFPE human pancreatic cancer using 1:10 dilution of Notch-1 antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) was performed using Aperio AT2 (Leica Biosystems). Cytoplasmic staining in epithelial cells was observed. Staining was performed by Histowiz.
Knockout Validated: Notch-1 Antibody (mN1A) [NB100-78486] - Lysates of HeLa human cervical epithelial carcinoma parental cell line and Notch-1 knockout (KO) HeLa cell line. PVDF membrane was probed with 2.0 ug/mL of Mouse Anti-Human Notch-1 Monoclonal Antibody (Catalog # NB100-78486) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog #HAF018). Specific band was detected for Notch-1 at approximately 110 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.
Flow Cytometry: Notch-1 Antibody (mN1A) [NB100-78486] - Intracellular flow cytometric staining of 1 x 10^6 CHO (A) and MCF-7 (B) cells using Notch1 antibody (dark blue). Isotype control shown in orange. An antibody concentration of 1 ug/1x10^6 cells was used.

BG01V human embryonic stem cells were fixed using formaldehyde, resuspended in lysis buffer, and sonicated to shear chromatin. Oct‑3/4/DNA complexes were immunoprecipitated using 5 μg Goat Anti-Human/Mouse Oct‑3/4 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1759) or control antibody (Catalog # AB-108-C) for 15 minutes in an ultrasonic bath, followed by Biotinylated Anti-Goat IgG Secondary Antibody (Catalog # BAF109). Immunocomplexes were captured using 50 μL of MagCellect Streptavidin Ferrofluid (Catalog # MAG999) and DNA was purified using chelating resin solution. The nanog promoter was detected by standard PCR.
Alkaline phosphatase (ALPL) and Oct‑3/4 were detected in human BG01V embryonic stem cells using 10 µg/mL Human/Mouse/Rat ALPL Monoclonal Antibody (Catalog # MAB1448) and 10 µg/mL Goat Anti-Human/Mouse Oct‑3/4 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1759). Cells were incubated with primary antibodies for 3 hours at room temperature. Cells were stained for ALPL using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (pseudo-stained green; Catalog # NL007), and stained for Oct-3/4 using the NorthernLights 637-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL002). Chromatin Immunoprecipitation (ChIP): csl/RBPJK Antibody [NBP1-33427] - Sheared, cross-linked chromatin was prepared from Jurkat T-ALL cells using the Covaris TruChIP kit and shearing with the LE220 sonicator. The chromatin buffer was adjusted to 150 mM NaCl, 20 mM Tris-Cl pH 8, 2 mM EDTA, and 1% Triton X-100. Three antibodies, as well as pure pre-immune rabbit IgG control, were added to each IP reaction, consisting of 7 million cells, and incubated overnight with mixing at 4 C. The antibodies were captured with Magna ChIP Protein A+G beads (Millipore), washed with three wash buffers, and the DNA eluted and purified with the iPure v2 kit (Diagenode). The purified DNA was normalized to the input control with qPCR quantification of the insulin exon 1 gene (INS) and an RBPJ binding site in the promoter of HES1. Image from verified customer review.

Immunocytochemistry/Immunofluorescence: EpCAM/TROP1 Antibody (AUA1) [NB600-1182] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton X-100. The cells were incubated with anti-EpCAM/TROP1 (AUA1) conjugated to Alexa Fluor 488 [NB600-1182AF488] at 10ug/ml for 1 hour at room temperature. Nuclei were counterstained with DAPI (blue). Cells were imaged using a 40X objective.
