Background

The protein encoded by this gene is a transcription factor that contains two GATA type zinc fingers. The encoded protein is known to bind to hepatocyte nuclear factor-1alpha (HNF 1 alpha), and this interaction is essential for cooperative activation of the intestinal lactase phlorizin hydrolase promoter. In other organisms, similar proteins may be involved in the establishment of cardiac smooth muscle cell diversity. GATA5 is induced at an early stage of endothelial-endocardial differentiation prior to expression of such early endocardial markers as Tie2 and ErbB3. It is required for differentiation of cardiogenic precursors into endothelial endocardial cells.

  1. We are interested in performing immunohistochemistry in human tissue with your anti-GATA5 antibody (NBP1-51215). We would like to know whether you have used this antibody in immunohistochemistry, to make sure that it works (we tried other antibodies that did not work). In addition, we also wanted to know whether it is a problem for the antibody that the tissue has been fixed in cold acetone.
    • We have validated this antibody on formalin fixed paraffin-embedded human breast carcinoma tissue section using its 1:100 dilution. I have uploaded the image on datasheet and it will appear online in next few hours. As you already know, acetone is a coagulant fixative while formalin fixes the tissues via cross-linking of proteins. In general, an antibody that work for formalin fixed tissues, works on all platforms of IHC as far as different fixatives are concerned. When working with acetone or methanol fixed tissues, the permeablization is often considered optional or done with mild detergent treatment (PBS with 0.05 - 0.1% Triton X100). To answer your main question, we strongly believe that NBP1-51215 will work on your acetone fixed sections but you will need to optimize your assay conditions to see what works the best for you.
