Fibronectin/Anastellin Antibody (DH1) Summary

The antibody reacts with Fibronectin.
Mesenchymal Cells Marker
The antibody is specific to extradomain A (EDA) sequence of a cellular fibronectin and recognizes thus only the cellular fibronectin. Mab DHI has been shown to specifically block chondrocyte condensation in chicken embryos (5). The antibody cross reacts with human, chicken, rat, rabbit and guinea pig. Fibronectins are diverse high molecular weight cell adhesion glycoproteins found in basement membranes, in the interstitial connective tissue matrix, and in soluble in plasma and other body fluids. In adult tissues most fibronectin is produced by hepatocytes (pFn) that differ from locally produced cellular fibronectins. A monoclonal antibody (BF12) has been produced to fibronectin that reacts with both the cellular and plasma forms of the protein and recognizes the cell-binding region in fibronectin. The monoclonal antibody DH1 recognizes only cellular form of Fibronectin. DH1 is specific to the extra domain A (EDA) of the molecule. cFns are found in some basement membranes during development and only in distinct tissues in adult, for instance in smooth muscle and alveolar walls of airways. cFns are abundant in the plasma of carcinoma patients and in the stroma of various carcinomas. Monoclonal antibody (Mab) to cFn is derived from the hybridoma produced by fusion between myeloma cells and Balb/c spleen cells. Purified fibronectin from A8387 fibrosarcoma cells was used as an immunogen (21).

Integrin beta1 also known as integrin VLA-4 beta chain, fibronectin receptor beta chain, and CD29) is a 130 kD integrin beta chain containing VWFA-like domains. Five isoforms have been reported (beta1A-1D, 1C-2). This Type I membrane protein is a subunit of the heterodimeric receptor for laminin, fibronectin, collagen, fibrinogen, VCAM1, cytotactin, osteopontin, epiligrin, thrombospondin, and vitronectin. Signal transduction through the integrins affects embryogenesis, hemostasis, tissue repair, immune response, and metastatic spread of tumor cells. Integrin beta1 can be upregulated by LPS with soluble CD14 and TNF-alpha; downregulation of integrin beta1 by alpha-melanocyte stimulating hormone and progesterone. Integrin beta1 is modified by phosphorylation and glycosylation. Integrin beta1 partners with integrin alpha subunits alphav, and alpha1 through alpha1.1. The Poly6004 antibody recognizes the N-terminal region of human integrin beta1 and has been shown to be useful for Western blotting. Partners with integrin alpha subunits alphav, alpha1 through alpha11.

FAQs for Fibronectin/Anastellin Antibody (NBP1-51723). (Showing 1 - 2 of 2 FAQ).

  1. One of our clients is looking for antibodies against tenascin C and fibronectin that could be used for FFPE rat tissues. These antibodies should exclusively recognize tenascin C and fibronectin and do not cross react with each other. Can you inform me if you have these antibodies?
    • Here is a link to Tenascin C antibodies we have validated in rat tissues for IHC-P Here are antibodies we have validated in fibronectin in rat tissues for IHC-P
  2. What is the dilution factor for IHC for starting? Because the range of dilution factor on the datasheet is so broad, I am confused which dilution factor is suitable for starting.
    • I can recommend the following dilution to start. Keep in mind that these will all vary greatly by sample type, so these are just suggestions. You may need to use more or less antibody. For catalog number NBP1-51723, the dilution range is 1:500-1:1000.
