Cytokeratin 18 Antibody (RGE 53) Summary

Hela cells.
Epithelial Marker
Keratin RGE 53 specifically recognizes glandular epithelial cells from gastro-intestinal tract, respiratory tract and urogenetical tract. As well as endocrine and exocrine tissues and mesothelial cells. It is unreactive with stratified squamous epithelia (esophagus, epidermis, ectocervix). In transitional epithelium of the bladder RGE53 only reacts with the superficial (umbrella) cells. RGE53 can distinguish adenocarcinomas and mesotheliomas from squamous cell carcinoma and non-epithelial tumours. The antibody is directed against 45 kD cytokeratin protein (cytokeratin 18). Cross reactivity with; Keratin from most mammals, birds. Immunogen: Hela cells.
Tissue culture supernatant

Application Notes
Frozen sections, Cytological material, flow cytometry and Western Blotting. Working dilutions: 1:5-1:10 (Based on the indirect method, with a normal ABC method the dilution can be 5-10 times higher, but the optimal dilution should be tested by serial dilution). Positive control: adeno carcinoma

Reactivity Notes

Mammal, Avian. Please note that this antibody is reactive to Mouse and derived from the same host, Mouse. Additional Mouse on Mouse blocking steps may be required for IHC and ICC experiments. Please contact Technical Support for more information.

Cytokeratin 18 (CK-18), a normal constituent of the hepatocyte cytoskeleton) is a standard tumor marker. CK-18 is elevated in patients with non-malignant liver diseases, particularly in those with prominent cytolysis (1). CK-18 and the homologous mouse Endo B are type I IF protein subunits whose expression is restricted in adults to a variety of simple epithelial tissues. Keratin 8 and 18 (CK8/18) are the major keratin pair in simple-type epithelia, as found in the liver, pancreas, and intestine. Transgenic mice that express mutant keratin 18 develop chronic hepatitis, and have an increased susceptibility to drug-induced hepatotoxicity (2). Studies shows that high CK18 expression in tumor cells is associated with reduced invasiveness in vitro and lack of tumorigenicity in nude mice. It has been shown that high K18 expression correlated with a good prognosis and that reducing K18 expression increased the aggressiveness of established breast cancer cell lines (3).
