Cytokeratin 1 Antibody Summary

Immunogen
keratin extracted from human skin. Antigen location, cytoplasma. cross reactive with keratin from most mammals and birds. In immunohistochemistry nearly all cultured epithelial cell are positive, all non-epthilial cells are negative.
Specificity
The polyclonal antibodies react with keratin of most mammals and birds. A positive reaction is obtained with all stratified squamous epithelia and in epidermal appendages such as hairfollicles and sebaceous glands. Also the epithelia of the urinary tract, stomach, small and large intestine, uterine endometrium, bile ducts, prostate, kidney collecting tubules, thymic reticulum cells, mesothelium and to a lesser extend hepatocytes, adenocarcinomas, trasitional cell carcinomas and anaplastic carcinomas can be identified by this antiserum. Renal cell adenocarcinomas and small cell anaplastic lung tumors may remain negative, when examined in paraffin sections. Hepatocytes and pancreatic acini show a positive staining. Immunogen: keratin extracted from human skin. Antigen location: cytoplasma. Cross reactive with keratin from most mammals and birds. Application Notes
The antibodies can be used to identify by means of immunohistochemistry mesotheliomas, squamous cell carcinomas, adenocarcinomas, transitional cell carcinomas and anaplastic carcinomas. Renal cell adenocarcinomas and small cell anaplastic lung tumors may remain negative, when examined in paraffin sections. For immunohistochemistry on frozen sections fixation for 3 minutes with 1% formaldehyde is recommended. For formaldehyde fixed paraffin embedded tissue sections proteolytic pretreatment with pronase or trypsine is recommended. Working dilution 1:50 (based on the indirect method, with a normal ABC method the dilution can be 5-10 times higher, bu the optimal dilution should be tested by serial dilution). Dilute immediately before use with PBS or Tris. Recommended positive control: Squamouse cell carcinoma, skin, cervix.

Background

Cytokeratins, a group of at least 29 different proteins, are characteristic of epithelial and trichocytic cells. Cytokeratins 4, 5, 6 and 8 are members of the type II neutral-to-basic subfamily. Cytokeratin peptide 4 (59 kDa) is the secondary type II keratin expressed in non cornified stratified squamous epithelia. Cytokeratin peptide 5 (58 kDa) is the primary type II keratin in stratified epithelia, while cytokeratin type 8 (52 kDa) is a major type II keratin in simple epithelia. Cytokeratin 6 (56 kDa) is a "hyperproliferation" cytokeratin expressed in tissues with natural or pathological high turnover. Cytokeratins 10, 13 and 18 are members of the type I acidic subfamily. Cytokeratin peptide 10 (56 kDa) is the secondary type I keratin expressed in cornified epithelia. Cytokeratin 13 (54 kDa) is the secondary type I keratin expressed in non-cornified stratified squamous epithelia. Cytokeratin 18 (45 kDa) is the primary type I keratin expressed in simple epithelial cells.
