Application Notes
This JJ319 antibody has been tested by flow cytometric analysis of rat splenocyte suspensions. This can be used at less than or equal to 1.0 ug per test. A test is defined as the amount (ug) of antibody that will stain a cell sample in a final volume of 100 uL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. The JJ319 antibody has also been reported for use in Immunoprecipitation. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

Background

The JJ319 monoclonal antibody reacts with the rat CD28 molecule, expressed by thymocytes, mature T cells, and a subset of NK cells. CD28 is a ligand for CD80 (B7-1) and CD86 (B7-2) and is a potent costimulator of T cells. Signaling through CD28 augments IL-2 and IL-2 receptor expression as well as cytotoxicity of CD3-activated T cells.
