Immunocytochemistry/Immunofluorescence: AHR Antibody [NB100-128] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti AHR [NB100-128] at a 1:100 dilution overnight at 4C and detected with an anti-goat DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunohistochemistry-Paraffin: AHR Antibody [NB100-128] - Analysis of FFPE human liver using 1:200 conc. of AHR antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Staining was performed by Histowiz.

PVDF membrane was probed with 0.5&amp;nbsp;µg/mL Human Phospho-p27/Kip1 (T157) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1555) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band for Phospho-p27/Kip1 (T157) was detected at approximately 27 kDa (as indicated). The phospho-specificity of this antibody was supported by decreased labeling following treatment with 600 U lambda -phosphatase ( lambda -PPase) for 1 hour. This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Goat Anti-Human/Mouse/Rat Growth Hormone Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1067) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog&amp;nbsp;# DY998) followed by Substrate Solution (Catalog&amp;nbsp;# DY999) and stopping the enzymatic reaction with Stop Solution (Catalog&amp;nbsp;# DY994).
Recombinant Human Growth Hormone (Catalog&amp;nbsp;# 1067-GH) stimulates proliferation in the Nb2‑11 rat lymphoma cell line in a dose-dependent manner (orange line). Immunohistochemistry-Paraffin: AIP/ARA9 Antibody (35-2) [NB100-127] - AIP/ARA9 was detected in immersion fixed paraffin-embedded sections of human spleen using Mouse Anti-Human AIP/ARA9 (35-2) Monoclonal Antibody (Catalog # NB100-127) at 1:300 for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte&#8482; HRP Polymer Antibody (Catalog # VC001). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to the cytoplasm in splenocytes.
