Background

This encoded protein is thought to be a secreted protein belonging to the disintegrin metalloproteinase family. Its expression is upregulated during dendritic cells maturation. This protein may play an important role in dendritic cell function and their interactions with germinal center T cells.

Immunocytochemistry/Immunofluorescence: ADAM28 Antibody [NBP2-67246] - U937 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-ADAM28 at 10 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Strong bands at 40, 45kDa correspond to DCX protein, detected only in the developing brain.
Immunocytochemistry/Immunofluorescence: Doublecortin Antibody (3E1) [NBP1-92684] - Analysis of cortical neuron-glial cell culture from E20 rat stained with mouse mAb to Doublecortin (DCX), NBP1-92684, dilution 1:1,000 in red, and costained with chicken pAb to Microtubule Associated Protein2 (MAP2), dilution 1:10,000 in green. The blue is DAPI staining of nuclear DNA. DCX antibody reveals strong cytoplasmic staining in a population of small developing neurons and their processes, while MAP2 antibody stains dendrites and perikarya of mature neurons. Therefore DCX antibody is an excellent marker of developing neuronal cells.
Immunocytochemistry/Immunofluorescence: Doublecortin Antibody (3E1) [NBP1-92684] - Rat brain neural cultures stained with NBP1-92684 (green), a chicken polyclonal antibody to GFAP (NB300-213, red) and DNA (blue). The NBP1-92684 antibody reveals strong cytoplasmic staining in a population of small developing neurons. These cells are often found in small clumps as in these cultures, as in this example. Note that they are not positive for MAP2, which is characteristic of more mature neurons, and that the mature neuron shown is negative for NBP1-92684. This Doublecortin antibody is therefore an excellent marker of developing neuronal cells.

Western Blot: TRAILR2/TNFRSF10B Antibody [NB100-56618] - TRAIL R2/TNFRSF10B Antibody [NB100-56618] - Total protein from HL-60, HepG2, HCT-116 and human placenta was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-TRAIL R2 in 1% non-fat milk in TBST and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.
Immunohistochemistry-Paraffin: TRAILR2/TNFRSF10B Antibody [NB100-56618] - TRAIL R2/TNFRSF10B Antibody [NB100-56618] - Analysis of a formalin fixed paraffin-embedded (FFPE) human brain cerebellum using 1:200 conc. of TRAIL R2/TNFRSF10B antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 30 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 9.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 15 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Cytoplasmic staining was observed in the Purkinje cell layer.
Immunohistochemistry: TRAILR2/TNFRSF10B Antibody [NB100-56618] - Pharmacodynamic assays for ONC201. IHC analyses of CHOP, TRAILR2/TNFRSF10B, and double-stranded DNA breaks (TUNEL) in baseline and ONC201-treated biopsies for an endometrial cancer patient. The biopsy was done 9.8 months after starting ONC201 treatment (7 days after the most recent ONC201 dose). In the presence of a cross-linking antibody, Mouse polyHistidine Monoclonal Antibody (Catalog # MAB050) and the metabolic inhibitor actinomycin D (1 µg/mL), Recombinant Human TRAIL R1/TNFRSF10A Fc Chimera (Catalog # 347‑DR) inhibits Recombinant Human TRAIL/TNFSF10 (Catalog # 375‑TL) induced cytotoxicity in the L‑929 mouse fibroblast cell line in a dose-dependent manner (orange line), as measured by crystal violet staining. Under these conditions, inhibition of Recombinant Human TRAIL/TNFSF10 (12 ng/mL) activity elicited by Recombinant Human TRAIL R1/TNFRSF10A Fc Chimera (10 ng/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human TRAIL R1/TNFRSF10A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF347). The ND50 is typically 0.02‑0.055 µg/mL.

Second antibody was biotinylated goat anti-mouse IgG at 23C for 1 hour. Development using avidin-biotin complex 23C for 1 hour plus 3,3-diaminobenzadine for 15 min and counterstained with haematoxylin.
