Background
Alzheimer's disease (AD) is a devastating neurodegenerative ailment which affects mainly the elderly people and leaves them with progressive intellectual deterioration involving memory, language, judgment, problem solving capability etc. and it is characterized by the accumulation of senile plaques (in brain tissue) composed of amyloid-beta (Abeta) peptides predominantly consisting of 40 (Abeta 40) and 42 (Abeta 42) amino acids which are generated via processing of APP (amyloid precursor protein). APP gets hydrolyzed by beta-secretase to generate a 99-residue membrane-associated C-terminus fragment (APP-C99) which is further cleaved to release a ~4-kDa peptide, Abeta, and AICD (APP intracellular domain) via an unusual form of proteolysis involving gamma-secretase mediated protein cleavage within the transmembrane domain (at residue +40 or +42). On the other hand, alpha-secretase cleaves APP in the middle of the abeta region resulting to the release of a large ectodomain (alpha-APPs), leaveing behind a C-terminus fragment of 83 amino acids (APP-C83) in the membrane which is further cleaved by gamma-secretase to generate p3, an N-terminally truncated form of abeta. The excessive accumulation of abeta peptides in AD is generally due to increased endoproteolytic cleavage of membrane bound APP, over-expression of APP and/or decreased clearance of abeta from the CNS.

FAQs for Abeta 40 Antibody (NB300-225). (Showing 1 - 1 of 1 FAQs).
What is the molecular weight and molar concentration of Abeta 40 Antibody (NB300-225)?
IgG molecules have 2 heavy (~55 kDa) and 2 light (~25 kDa) chains, and so the size of an IgG is 55 kDa x 2 + 25 kDa x 2 = ~160 kDa. Also all IgG molecules have the similar sizes on PAGE gels. Regarding the concentration, because this antibody is unpurified rabbit antiserum, we don't know the concentration and apologize for any inconvenience. We provide recommended dilutions for each validated application (Western Blot 1:500-1:1000, Immunocytochemistry/Immunofluorescence 1:1000, and Immunoprecipitation 1:100), and have a 100% guarantee for verified applications and species
The C99 fragment of amyloid precursor protein (APP) Alzheimer’s Disease (AD) is a neurodegenerative disorder that is characterized by an abundance of the beta-amyloid peptide in the brain. When AD was first discovered, it was determined that beta-amyloid was produced as a result of the prote... Read full blog post.
