Immunocytochemistry/Immunofluorescence: MHC Class I Antibody (OX18) [NB120-6405] - Major histocompatibility complex (MHC) I and II as well as Transporter associated with antigen presentation II (TAPII) were analyzed, using immunocytochemistry on rat Schwann cells (SCs). Corresponding merges are shown in the bottom rows. Treatment of SCs with IL-17 was performed at concentrations of 0.5 and 50 ng/mL. Graphs to the right show densitometry quantification. SCs showed expression of MHCI > TAPII > MHCII, which increased after IL-17 treatment. MHCI was mainly detected in the cytoplasm and the expression increased in a dose-dependent manner after IL-17 treatment, significant for 0.5 ng/mL and 50 ng/mL (**P <=0.01). Image collected and cropped by CiteAb from the following publication (http://jneuroinflammation.biomedcentral.com/articles/10.1186/1742-2094-11-63), licensed under a CC-BY license.Immunohistochemistry-Paraffin: MHC Class I Antibody (OX18) [NB120-6405] - Analysis of FFPE rat brain cerebellum using MHC Class I (OK18) antibody at 1:200 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Endothelial staining was observed. Staining was performed by Histowiz.

Western Blot: Doublecortin Antibody (3E1) [NBP1-92684] - Analysis of rat whole brain lysates using mouse mAb to Doublecortin (DCX), NBP1-92684, dilution 1:1,000 in green: [1] protein standard (red), [2] adult rat brain, [3] embryonic E20 rat brain. Strong bands at 40, 45kDa correspond to DCX protein, detected only in the developing brain.Immunocytochemistry/Immunofluorescence: Doublecortin Antibody (3E1) [NBP1-92684] - Analysis of cortical neuron-glial cell culture from E20 rat stained with mouse mAb to Doublecortin (DCX), NBP1-92684, dilution 1:1,000 in red, and costained with chicken pAb to Microtubule Associated Protein2 (MAP2), dilution 1:10,000 in green. The blue is DAPI staining of nuclear DNA. DCX antibody reveals strong cytoplasmic staining in a population of small developing neurons and their processes, while MAP2 antibody stains dendrites and perikarya of mature neurons. Therefore DCX antibody is an excellent marker of developing neuronal cells.

Western Blot: GRAIL/RNF128 Antibody [NBP2-24610] - Bone marrow progenitors were isolated from C57BL/6 mice and incubated with 20ng/ml GM-CSF. (a) Cells were collected at days 0-9. A portion of the cells at day 8 were stimulated with OVA for 24h. Collected cells were processed for protein extraction followed by immunoblot analysis with antibodies to the indicated proteins. Blots were stripped of antibodies prior to probing with the next one. The two braces on the left represent two different gels of the same samples. Signals were quantified and are shown as values under the respective blots. Knockout Validated: B7-2/CD86 Antibody (BU63) [NBP2-25208] - Western blot shows lysates of Ramos human Burkitt's lymphoma parental cell line and B7-2 knockout (KO) Ramos cell line. PVDF membrane was probed with 1.0 ug/mL of Mouse Anti-Human B7-2 Monoclonal Antibody (Catalog # NBP2-25208) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog #HAF018). Specific band was detected for B7-2 at approximately 90 kDa (as indicated) in the parental Ramos cell line, but is not detectable in the knockout Ramos cell line. This experiment was conducted under reducing conditions.Immunohistochemistry: B7-2/CD86 Antibody (BU63) [NBP2-25208] - Macrophage polarization states. Distribution and proportion of CD86+ macrophages (brown) were identified using IHC procedures. Intensity for the color response to antibodies was visible in lung tissues from all groups. Population proportions for the stained cells as shown by red arrows were calculated as a fold change of the control. (d). The results were expressed as Mean +/- SD (n = 3). **: a p-value of < 0.01 vs NS, OVA/BUD and OVA/PCI except the number of CD86+ cells in OVA/PCI group. #: P < 0.05 vs NS and ##; P < 0.01 vs either NS or OVA/BUD group.
