Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Phycoerythrin.

Western Blot: Osteoprotegerin/TNFRSF11B Antibody (98A1071) [NB100-56505] - Analysis of Daudi lysate using the OPG antibody at 1 ug/ml.Immunocytochemistry/Immunofluorescence: Osteoprotegerin/TNFRSF11B Antibody (98A1071) [NB100-56505] - MG-63 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100.  The cells were incubated with anti-Osteoprotegerin (98A1071) at 5 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution.  Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution.  Nuclei were counterstained with DAPI (Blue).  Cells were imaged using a 40X objective.

Lane 2: Serum-starved HeLa cells were stimulated with 20% FBS (fetal bovine serum) for 2 hours. NBP2-50037 recognizes bands in the range of 50-65 kDa, which represent multiple forms of c-Fos. Serum starvation attenuates c-Fos expression, while 20% FBS strongly stimulates c-Fos expression. Functional (Inhibition): NFkB p105/p50 Inhibitor Peptide Set [NBP2-29323] - TLR5/NF-kB/SEAPorter HEK 293 (NBP2-26277) cells were plated in 96-well plates at 5 x 10^4 cells/well for 16 h. Cells were preincubated with different concentrations (0, 1, 5, 10, 25 and 50 uM) of Inhibitory Peptide (NBP2-29323) and Control Peptide (NBP2-29334) for 1 h. Cells were then stimulated with 1 ng/ml Flagellin (NBP2-25289) for 24 h. Secreted alkaline phosphatase (SEAP) was analyzed using SEAPorter Assay Kit (NBP2-25285). *p < 0.05 versus control peptide at the corresponding concentrations (Mann-Whitney U test).

TRAF6 Inhibitor Peptide Set [NBP2-26506] - The TRAF6 inhibitory peptide inhibits osteoclast ifferentiation: RAW264.7 cells were plated on 24-well plates and stimulated with RANKL and M-CSF and incubated in the presence of different concentrations of control peptide (PTD) or the TRAF6. The TRAF6 at the concentration above 30 uM suppressed osteoclast differentiation.
