Immunocytochemistry/Immunofluorescence: GM130/GOLGA2 Antibody [NBP2-53420] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with anti-GOLGA2 at a 1:200 dilution overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Western Blot: ATF6 Antibody (70B1413.1) [NBP1-40256] - Image of anti-ATF6.  Whole cell protein from HeLa cells treated with and without 10 mM DTT was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST.  The membrane was probed with 5 ug/ml anti-ATF6 in 1% milk, and detected with an anti-mouse HRP secondary antibody using a maximum sensitivity ECL reagent.  Note the loss of the 90 kDa modified ATF6 protein (arrowhead) upon DTT treatment to activate the unfolded protein response.Immunocytochemistry/Immunofluorescence: ATF6 Antibody (70B1413.1) [NBP1-40256] - Untreated HeLa cells were fixed in -20C methanol for 10 min, air dried and rehydrated in PBS at room temperature for 5 minutes.  Cells were incubated with anti-ATF6 (1:20) for one hour at room temperature.  ATF6 reactivity (green) was detected with anti-mouse Dylight-488 secondary antibody.   Nuclei were counterstained with DAPI (blue).  Note the ER localization of ATF6.
