The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Cytoplasmic staining in the squamous epithelium was observed. Staining was performed by Histowiz.

Western Blot: NOD2 Antibody (2D9) [NB100-524] - HCMV infection induces NOD2 mRNA and protein in HFFs and U373 cells. E. U373 glioma cells were infected with HCMV Towne strain and levels of NOD1, NOD2 and GAPDH mRNAs were measured by qRT-PCR at indicated time points. F. HFFs were infected with HCMV (Towne) at MOI of 1 PFU/cell and levels of NOD2 protein and B-actin were determined 48 and 72 hpi. G. HFFs were infected with HCMV (Towne) strain at MOI of 0.03 or 3 PFU/cell and levels of NOD2 protein and B-actin were determined at 48 hpi. Quantitative data represent mean values (+/-SD) of triplicate determinations from three independent experiments (*p<0.05, **p<0.01, ***p<0.001, one-way ANOVA test). Image collected and cropped by CiteAb from the following publication (//doi.org/10.1371/journal.pone.0092704.g001) licensed under a CC-BY licence.Western Blot: NOD2 Antibody (2D9) [NB100-524] - Total protein from human THP1 and US-OS cells and mouse Raw264.7 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-NOD2 in 1% non-fat milk in TBST and detected with an anti-mouse HRP secondary antibody using chemiluminescence.

Simple Western: Caspase-1 Antibody (14F468) [NB100-56565] - Simple Western lane view shows a specific band for Caspase 1 in 1.0 mg/ml of HeLa lysate. This experiment was performed under reducing conditions using the 12-230 kDa separation system.Immunohistochemistry: Caspase-1 Antibody (14F468) [NB100-56565] - Activity of Casp1 in OHT-injured and normotensive control eyes. (A) Casp1 was detected by intraocular injection FLICA660-labeled substrate (green) in vivo 24 h after injury. Bright labeling (arrows) is evident in cells in the GCL and inner nuclear layer (INL) layers of the OHT-challenged retinas, a diffuse labeling of cell processes located in the IPL. Casp1 activity is diminished in Panx1-/- (Px1-/- OHT) retinas and WT retinas treated with probenecid (WT/Pbcd) at 12 h postinjury. Western Blot: NALP12 Antibody [NBP1-76293] - B6 mice (n = 6) were infected ocularly with 2.5 x 105 pfu/eye of HSV-1 laboratory strains McKrae, 17, F, RE or KOS or with clinical isolates KOS79 and KOS63 (mock-infected mice (n = 6) used as controls). 48 hrs later, animals were euthanized & corneas were excised & homogenized in lysis buffer, and WB was performed for NLRP3, NLRP6, NLRP12, IFI16/p204, & AIM2 (beta-actin used as a control). Three WB performed using different corneal lysates harvested from three sets of corneas from 2 mice (4 corneas/lysate). Graphs (mean plus SD) show fold change in expression of NLRP3, NLRP6, NLRP12, IFI16/p204, and AIM2 at 48 hrs post-infection compared to controls, and normalized to beta-actin. Representative densitometric analysis is shown compared to control.
