Description

Glutamate transporters (GLTs) inactivate the synaptically released glutamate thus preventing excessive neurotoxic stimulation of glutamate receptors and GLT1 is a predominant transporter in brain (responsible for up to 90% of extracellular glutamate clearance) that has two major isoforms, GLT1a (Slc2a1, transcript variant 1) and GLT1b (Slc2a1, transcript variant 2). Splice variant GLT1b contains 11 unique amino acids at C-terminus that makes it capable of interacting with PDZ domains of a number of proteins. Initially believed to be the neuronal isoform of GLT1, GLT1b now has been shown to localize to astrocytes in the normal brain and to neurons in pathological conditions. GLT1b forms co-immunoprecipitable complexes with PSD-95 in solubilizated rat brain extracts, complexes that also contained NMDA receptors, and GLT1b/PSD-95 interaction have been proposed to anchor GLT1b transporter close to the post-synaptic glutamate receptors, thereby permitting the fine regulation of glutamate concentrations in this microenvironment.
