Novus Biologicals introduces the new FasL-Strep Apoptosis Assay kits from IBA GmbH. A new patented fusion technology linking FasL with a “T4-foldon” globular protein provides for a superior bioactive, stable, well defined trimer. This Modified FasL is fused with a Strep tag making it ideal for apoptosis induction and detection. The advantages of the FasL-Strep are the defined trimeric structure, high stability, efficient apoptosis induction, and easy handling for detection, immobilization, and isolation via the strep-tag. Human FasL-Strep (CD95L-T4) is an engineered human Fas (CD95 ligand. C-Terminal fusion of the T-4-FOLDON to the extracellular FasL (CD95L) receptor-binding domain (E142-L281) results in bioactive, stable, well defined trimers in contrast to commonly used N-terminal fused coiled-coil structures. Furthermore, fusion of a Strep-tag II C-terminal to the T4-FOLDON motif provides downstream applications like in solution cross-linking of the trimeric Fas (CD95) ligand through the monoclonal antibody StrepMAB-Immo (Cat. No. 2-1517-001) for the intensification of apoptotic activity or immobilization on Strep-Tactin coated microplates (2-1501-001) for performing a geterogeneous apoptotic assay. Recombinantly extressed and purified from HEK293 cells. In the heterogeneous apoptosis assay format, the FasL-Strep molecule is immobilized on Strep-Tactin(R) (the cognitive receptor of Strep-tag(R)) coated microplates. After addition of the cells to be investigated, caspase 3/7 activity is determined for measuring extent of apoptosis. The FasL-Strep apoptosis assay described above is robust and reproducible due to the high stability of the FasL-Strep reagent. The test is easy to perform since common reagents are applied for read out. These properties identify this assay as an ideal basis for HTS assays for screening anti-apoptotic molecules.
