Much of the research into the causes of lung cancer have focused on the inactivation of tumor suppressor genes, activation of oncogenes, and/or stimulation of growth factors. Many tumor suppressor genes have been studied and shown to be involved in the development and progression of lung cancer. In addition, gene rearrangements (ROS1, RET) and gene fusions (EML4-ALK, CD74-NRG1) have proven to be important targets of therapy in a subset of lung cancer patients. Vascular endothelial growth factor (VEGF) has been extensively studied as a major element in lung cancer tumor development and metastasis. It is often overexpressed as lung cancers progress, strongly associated with increased microvessel density, and linked to decreased survival.
