<b>Orthogonal Strategies Validation. </b>Formalin-fixed paraffin-embedded tissue sections of human placenta were probed for HIF-2 alpha/EPAS1 mRNA (ACD RNAScope Probe, catalog #410598; Fast Red chromogen, ACD catalog # 322750). Adjacent tissue section was processed for immunohistochemistry using Rabbit Polyclonal (Novus Biologicals catalog # NB100-122) at 1:100 dilution with one-hour incubation at room temperature followed by incubation with anti-rabbit IgG VisUCyte HRP Polymer Antibody (Catalog # VC003) and DAB chromogen (yellow-brown). Tissue was counterstained with hematoxylin (blue). Specific staining was localized to trophoblastic cells.

An intracellular stain was performed on U-87 MG Cells with NB100-417 and a matched isotype control. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG APC-conjugated Secondary Antibody, (R&D Systems, F0111).

A431 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-Carbonic Anhydrase IX/CA9 Antibody NB100-417 at 2 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

An intracellular stain was performed on A431 cells with Carbonic Anhydrase IX/CA9 Antibody NB100-417 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (SA5-10033, Thermo Fisher).

IHC analysis of a FFPE tissue section of human breast cancer using CAIX antibody at 1:1000 dilution. The primary antibody bound to CAIX antigens in the tissue section was detected using a HRP labeled secondary antibody and DAB reagent. Nuclei of the cells were counterstained with hematoxylin. This CAIX antibody generated an expected cytoplasmic staining of CAIX protein with an intense signal around the cellular membranes in tumor cores. The latter are more likely to be hypoxic in growing tumors which signifies that the observed CAIX staining is specific.

Beclin 1 was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line using Rabbit anti-Beclin 1 Affinity Purified Polyclonal Antibody conjugated to FITC (Catalog # NB500-249F) (green) at 10 µg/mL overnight at 4C. U87 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti- NB100-664 at 1 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunohistochemistry-Paraffin: Beclin 1 Antibody [NB110-87318] - Analysis of a FFPE tissue section of human breast cancer using Beclin 1 antibody (Lot 71917) at 1:300 dilution. The primary antibody bound to Beclin 1 antigen in the tissue section was detected using a HRP labeled secondary antibody and DAB reagent. Nuclei of the cells were counterstained with hematoxylin. Cytoplasmic staining can be seen in the cancer as well as the stromal cells. The signal was strongest in a subset of stromal cells which appears to be the cancer associated fibroblasts.

Western Blot: Beclin 1 Antibody [NB110-87318] - Basal levels of autophagy in the hypothalamus of BACHD mice. The expression levels of selected autophagy markers were analyzed by Western blot in hypothalamic tissue from BACHD mice at 2, 6 and 12 months of age and compared with gender- and age-matched controls (n=3-6/group). The data showed a substantial maintenance of the autophagy flux, despite the presence of a metabolic phenotype in the BACHD mice already at early time points. VEGF, NB100-648, was used at a dilution of 1:1000 in TBS Tween and 5% milk powder. The protein was labeled at 40 kDa. Quantification of VEGFA in bone graft in the tibia of intrailiac cellularized goats at different times compared with tibial autograft and untreated bone defect. Evaluation of pre-cellularized graft in iliac crest in the treatment of bone defects in the tibia of goats. G1: cellularized graft for 4 weeks applied to tibial bone defect, G2: cellularized graft for 6 weeks applied to tibial bone defect, G3: cellularized graft for 8 weeks applied to tibial bone defect, G4: tibial autograft and G5: untreated bone defect. The sections of histological sections of goat fragments were deparaffinized, peroxidase blocked in methanol and 30% hydrogen peroxide (9:1), and antigen retrieval was performed with porcine gastric pepsin and HCl at pH 3.0 in an oven at 37°C for 30 minutes. The cells were incubated with Carbonic Anhydrase IX/CA9 Antibody [2D3] (NBP1-51691) at 2ug/ml overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.
