This Ku80 (5D5.8) antibody is useful for Immunocytochemistry/Immunofluorescence and Western Blot analysis where a band is seen at approx. 85 kDa. *The investigator should determine the optimal working dilution for a specific application.

Background

Heterodimeric Ku antigen (heterodimer of Ku70 and Ku80 subunits) is DNA double strand's molecular detector and it activates DNA protein kinase via direct binding in non-sequence-specific manner to free DNA termini. Encoded by XRCC5, Ku80 exist as heterodimer that associates with PRKDC in a DNA-dependent manner to form DNA-PK (DNA-dependent protein kinase) complex and binds with LIG4-XRCC4 complex also. The heterodimer also associates with NAA15 to create complex with tendency to bind DNA towards OCFRE (osteocalcin FGF response element) and also interacts with MSX2, RUNX2, ELF3, APLF, APEX1 etc. In DSBs repair, Ku80 plays its critical role in maintaining genome stability through non-homologous end joining (NHEJ) pathway mediated repair of eukaryotic DSBs which, if left unrepaired, may lead to chromosomal aberration, mutation or carcinogenesis. Ku80 involves in telomere maintenance, apoptosis, tumor suppression etc. and has been proposed to be a multifunctional caretaker with ability to suppress chromosomal aberrations and malignant transformation. Overexpression of Ku80 is associated with aggressive cancers such as bladder, cervical, pancreatic, gastric, breast and lung cancer, and is involved in drug resistance in case of ovarian cancer and leukemia. Individuals with SLE and related disorders also produce extremely large amounts of autoantibodies to Ku80.
