Featured Neuroscience Antibodies: Parkinson’s Disease Targets Approximately one million Americans suffer from Parkinson’s Disease, with 60,000 new cases expected to be diagnosed currently each year. Parkinson’s Disease belongs to a group of progressive neurodegenerative diseases know as synucleinopathies characterized by the presence of abnormal α-synuclein-immunoreactive inclusions within neurons.
Nitrated alpha-Synuclein is a soluble protein primarily localized at the presynaptic regions of axons. It can form filamentous aggregates, which are the major non- amyloid component of intracellular inclusions in many neurodegenerative synucleinopathies. Nitrated alpha-Synuclein has been found to reduce neuronal responsiveness to several apoptotic stimuli which leads to a decreased caspase-3 activation.
Leucine-rich repeat kinase 2 (LRRK2) is present largely in the cytoplasm but also associates with the mitochondrial outer membrane. Mutations in this gene have been associated with Parkinson's Disease 8 (PARK8). PARK8 is an autosomal-dominant late-onset parkinsonism with slow progression.
Four chromosomal loci (PARK2, PARK6, PARK7, and PARK9) associated with autosomal-recessive, early-onset parkinsonism are known. Positional cloning within the refined PARK7 critical region recently identified mutations in the DJ-1 gene in PARK7-linked families. Mutations in the PARK7/DJ-1 gene cause autosomal-recessive Parkinson's Disease.
