Spotlight on 53BP1
P53-binding protein 1 (53BP1) is involved in the early stages of cellular response to DNA double-strand breaks.  Upon DNA damage, 53BP1 is phosphorylated and relocalizes to the double-strand break sites. Methylation of histone H4 at Lys-20 is required for efficient localization to these sites.  53BP1 plays a redundant role in activation  of the downstream checkpoint effector proteins Brca1 and Chk2, but is required for the formation of Brca1 foci for the recruitment of repair/signaling proteins to sites of DNA damage. 53BP1 also plays a critical role in tumor suppression and is a putative substrate of ATM kinase. Cytochrome c

This antibody was given four out of five stars in a customer review submitted on May 27th, 2009.  The customer used the antibody for Immunofluorescent staining of mouse B cells at a 1:500 dilution.   The cells were first fixed with Formaldehyde and permeabilized using Triton-X100.  The 53BP1 antibody was incubated for one hour at room temperature in PBS + 0.2% BSA + 0.1% Tween 20.  The secondary antibody used was an Alexa fluor 633 goat anti-rabbit used at a dilution of 1:500, and was incubated at room temperature for one hour in PBS + 0.2% BSA + 0.1% Tween 20.
