Background

Parathyroid hormone (PTH), which is also designated parathyrin, is an 84 amino acid single chain peptide that functions to regulate calcium metabolism by raising blood levels of calcium through various mechanisms. PTH stimulates bone formation to increase bone mass and strength in rats and humans. Within the PTH molecule, the essential activity is associated with the first 34 amino acids at the amino-terminus of the molecule. The gene encoding PTH maps to human chromosome 11p15.3-p15.1. Parathyroid hormone-related protein (PTH-rP) is an autocrine factor that is structurally related to PTH yet, unlike PTH, which is synthesized only by the parathyroid cells, PTH-rP is synthesized by several cell types. PTH-rP regulates endochondral bone development and epithelial-mesenchymal interactions during the formation of the mammary glands and teeth. Isolated from the culture medium of a human lung cancer cell line, PTH-rP produces PTH-like effects that are characterized as humoral hypercalcemia of malignancy.

Western blot shows lysate of human liver tissue. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human Serum Albumin Monoclonal Antibody (Catalog # MAB1455) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for Albumin at approximately 65-70 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.    Simple  Western lane view shows lysates of human liver tissue, loaded at  0.2 mg/mL. A specific band was detected for Albumin at approximately  64 kDa (as indicated) using 10 µg/mL of Mouse Anti-Human  Serum Albumin Monoclonal Antibody (Catalog # MAB1455). This experiment was  conducted under reducing conditions and using the 12-230 kDa  separation system.         Albumin was detected in immersion fixed BG01V human embryonic stem cells differentiated to hepatocytes using Mouse Anti-Human Serum Albumin Monoclonal Antibody (Catalog # MAB1455) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. Cells were co-stained using Sheep Anti-Human CEBP alpha (Catalog # AF7094) and NorthernLights™ 493-conjugated Anti-Sheep IgG Secondary Antibody (green, Catalog # NL012).
