Nanog is one of the molecular markers suitable for recognizing the undifferentiated state of stem cells in the mouse and human. It is a divergent homeodomain protein that directs propagation of undifferentiated ES cells. Nanog mRNA is present in pluripotent mouse and human cell lines, and absent from differentiated cells. Nanog-deficient ES cells lose pluripotency and differentiate into extraembryonic endoderm lineage.

  Western  blot shows lysates of BG01V human embryonic stem cells, NTera‑2  human testicular embryonic carcinoma cell line, and D3 mouse embryonic stem  cell line. PVDF membrane was probed with 0.2 µg/mL of Recombinant Mouse  Anti-Human/Mouse SOX2 Monoclonal Antibody (Catalog # MAB2018R) followed by  HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018).  A specific band was detected for SOX2 at approximately 37 kDa (as  indicated). This experiment was conducted under reducing conditions and using  Immunoblot  Buffer Group 1.NTera‑2  human testicular embryonic carcinoma cell line was stained with Recombinant Mouse  Anti-Human/Mouse SOX2 Monoclonal Antibody (Catalog # MAB2018R, filled  histogram) or isotype control antibody (Catalog # MAB003,  open histogram), followed by Phycoerythrin-conjugated Anti-Mouse IgG  Secondary Antibody (Catalog # F0102B).  To facilitate intracellular staining, cells were fixed with Flow Cytometry  Fixation Buffer (Catalog # FC004)  and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I  (Catalog # FC005).    View our  protocol for Staining  Intracellular Molecules.

A specific band was detected for STAT3 at approximately 90 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.STAT3 was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Mouse Anti-Human/Mouse/Rat STAT3 Monoclonal Antibody (Catalog # MAB1799) at 3 µg/mL for 3 hours at room temperature.
