Background: Mouse IgG1 myeloma cells were grown in mice and ascitic fluids collected. The IgG1 was purified by a combination of salt fractionation, ion exchange and protein A chromatography. Protein concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG. By immunoelectrophoresis the IgG1 was shown to 1) react with antiserum specific for mouse IgG1, 2) not react with antiserum specific for other immunoglobulins, and 3) produce a single precipitation arc with antiserum against mouse serum identical to that produced with anti-IgG1.
