Western Blot: Histone H3 [ac Lys4] Antibody [NB21-1024] - Histone H3 [ac  Lys4] Antibody [NB21-1024] - WB analysis of Histone H3 K4ac in 1. HeLa histone prep and 2. NIH 3T3 histone prep lysates.
Immunocytochemistry: Histone H3 [ac Lys4] Antibody [NB21-1024] - Immunofluorescence: Histone H3 [ac  Lys4] Antibody [NB21-1024] - Histone H3 K4ac1 antibody was tested in HeLa cells with Dylight 488 (green). Nuclei and alpha-tubulin were counterstained with DAPI (blue) and Dylight 550 (red).
Chromatin Immunoprecipitation: Histone H3 [ac Lys4] Antibody [NB21-1024] - Histone H3 [ac  Lys4] Antibody [NB21-1024] - Chromatin from one million formaldehyde cross-linked Hela cells was used with 2ug of NB21-1024 and 20ul of magnetic IgG beads per immunoprecipitation. A no antibody (No Ab) control was also used. Immunoprecipitated DNA was quantified using quantitative real-time PCR and SYBR green dye, then normalized to the non-precipitated input chromatin, which is equal to one.

In transcription, H3K4Ac is prevalent at the promoters of active genes. Usually, this modification peaks in the cell cycle after dimethylation of H3K9 occurs, which indicates that there are sequential actions of these two modifications. Methylation that occurs on H3K4 concurrently with acetylation seems to act as an adjuster to the activation effects of acetylation. Shugoshin protein cannot bind to the centromere of active cells when H3K4 is acetylated, which reduces dimethylation, and thus slows meiosis and mitosis. Usually, H3K4Ac is a transitional modification, and will become further modified with methylation as transcription progresses, indicating complex transcriptional regulation.

  1. In the immunohistochemistry paraffin section protocol....the PBS buffer....should it be at certain pH?
    • For IHC application, different labs cites the pH of PBS buffer in the range of 7.1 - 7.6 but most commonly used pH is pH 7.4 (this is what we use in our lab). You may use this PBS (pH 7.4) for making permeablization buffer, antibody diluent buffers and for making wash buffer. For more on the protocol that we use in our lab and for IHC-P troubleshooting suggestions, you may visit: IHC-P Protocol and IHC-P Troubleshooting.
  2. I was wondering what does Format 7C mean as far as anitbodies go?
    • 7C is a fluorophore - it is useful for FLOW. The details of the fluor are (A=425, E=500)
